Cytometry

This laboratory gives technical support to those users who are interested in its areas.

  • Study of technical viability and training in the use of flow cytometers and data analysis programs.
  • Cellular cycle analysis in fresh, cryopreserved or in-paraffin samples by means of flow cytometry.
  • Antigen proliferation and cellular cycle biparamedical study by means of flow cytometry.
  • Multi paramedic study of cell populations by means of flow cytometry. Immunophenotyping.

  • Sorting.
  • Interphase or metaphase FISH analysis with one or two specific probes.

Sequencing

The existing equipment in the Sequencing Laboratory is the most advanced that is commercialised, and it allows for various approximations to the DNA analysis of discreet organisms.

Two methodologies can be applied: DNA sequencing and genotyping by means of the PCR-generated fragment size analysis (AFLPs, SNP's, SSCP's, Microsatellites, etc.).

Technical support

The unit has expert personnel in the fields such as sequencing and development and use of molecular markers in population studies as well as identification of species.

The cooperation of said personnel is focused on the technical viability of the different ideas proposed by the users as well as advising on the design and selection of markers or the interpretation of the results obtained.

This cooperation is free-of-charge, and its only goal is to allow the access to this techniques to as many users as possible; therefore, allowing the integration of these techniques in their fields.

DNA sequencing

By automatic sequencing the nucleotidic sequence of a DNA sample can be obtained. This can be cloned in a vector or have been amplified by PCR.

Within the Unit, there are two data analysis systems available which allow obtaining reads up to 550 and 950 bases respectively.

To do the sequencing, the most advanced chemistry is used (BigDye(r) Terminator v3.1). It offers improved performance over other chemistries in the sequencing of difficult samples. Besides, this chemistry generates data with an even peak height which ensures a more precise base assignation to detect mutations or heterozygotes.

Every sample is purified by means of size exclusion columns before being analysed with the purpose of deleting the background noise as well as the leftover terminators. This helps in the obtention of samples with an even peak height, giving, as a result, a more precise assignation. It is advised for non-expert users to consult the sample preparation guide

DNA Genotyping

By genotyping techniques, it can be seen one or various DNA fragments simultaneously amplified. This way, band patterns which can be specific for each species or that contain variability between the individuals of a certain species are obtained.

Within the methodologies which provide this genotyping, the following uses of microsatellites can be underlined: AFLP's, SNP's and SSCP's. All of them are susceptible of being analysed by means of the Sequencing Unit equipment.

Fields of application

Within the fields of application, the most relevant uses for the techniques are the following:

  • Basic research in molecular genetics: Phylogenetic and microorganism evolution study.
  • Basic research in Molecular Ecology: Population studies, larval-state species identification, individual genetic marker for field studies, etc.
  • Biomedicine: Molecular characterisation of clinically relevant genes, genetic predisposition to some cancer types, oncogenic mutation detention, etc.
  • Human genetics: Hereditary illnesses diagnostics. Genetic advice.
  • Legal medicine: Identification of individuals, paternity test.
  • Microbiology: Bacterial species identification, virus identification.
  • Vet: Pedigrees, gene identification in improvement programs, endangered species preservation, bird sexing, etc.
  • Food processing: Vegetable variety identification, traceability, fraud detection (species identification in the sample).

From a small feather or blood sample, enough DNA can be extracted to identify their sex in a 100% reliable way.

The technique is based in the PCR amplification of some DNA areas of interest: one is located in the Z chromosome (both present in the male and female) and the other is located in the W chromosome (which is exclusive of females). Then, a capillary electrophoresis system for the detection of the obtained products is used. In the case of males, a single product belonging to the chromosome Z (ZZ) would be obtained, and, in the case of females, two products would be obtained, one belonging to the chromosome Z and the other belonging to the chromosome W (ZW).

The result is obtained two days after the samples have been handed in. If the user wants the samples to be shipped, they must contact the Unit personnel.

The sexing price is of 15 € per specimen.

The services offered by this Unit are the following:

  • Creation of rabbit polyclonal antiserum as well as purification of the lgG fraction.

  • Cellular line cultures as well as the possibility of using the culture room by authorised personnel.
  • Advise and creation of the design and analysis of cytotoxicity as well as cell proliferation.

  • Cell culture contamination analysis by means of Mycoplasma.
  • Protein purification by means of ÄKTA FPLC chromatography.
  • Cryopreservation of biological samples.
  • DNA, RNA and protein quality and quantity analysis by means of the Agilent Bioanalyzer 2100 equipment.
  • Advice and technical support with biomolecular techniques (biomolecular extraction, electrophoresis, and immuno-tests).
